LONDON (AP) -Manchester City has plenty of additional motivation to beat Sunderland in the Premier League on Saturday.

Victory would put leader Manchester United under pressure, pay Sunderland back for what was only its second Premier League defeat of the season and give one player the pleasure of beating his hometown club.

Sunderland-born England winger Adam Johnson played the entire game at the Stadium of Light in January, so he was on the field to see Ji Dong-won score a last-minute winner in a match City had dominated.

"That defeat was hard to take because we played really well, dominated the match but the ball just wouldn't go in the net for us," Johnson said. "I don't think they had any shots of note during the game and then they nick it right at the death, so that's certainly in the back of our minds going into this game."

With defending champion United not in action until it plays Blackburn on Monday, City can draw level on points at the top of the table if it beats the Black Cats.

City slipped slightly further behind United last weekend with a 1-1 draw against Stoke but has won a Premier League-record 20 straight games at home and is a strong favorite to beat a side resurgent under manager Martin O'Neill.

"I'm sure they won't be looking forward to coming to our ground," said Johnson, who is close friends with Sunderland midfielder Lee Cattermole. "We've been fantastic on our own patch and we feel we can beat anyone, so we'll be looking to make it 21 Premier League wins in a row against them."

While Johnson is in contention to start, City striker Sergio Aguero is out with a foot injury. Manager Roberto Mancini would not say what the injury was and raised the prospect that it was self-inflicted or sustained in an accident by stating only that it was "stupid."

Aguero's absence could give Carlos Tevez the chance to start a match for the first time since Sept. 10. The Argentina striker has made three substitute appearances since returning from an unsanctioned six-month break.

United's game against Blackburn is the first of four in a row against teams in the bottom six of the Premier League and manager Alex Ferguson said it is crucial his team takes advantage to stay ahead of City until the teams meet on April 30.

"We have a small advantage: it's not a great advantage; it can be changed in one game," Ferguson said. "We just have to take each game as it comes along. It's the only thing we can do."

Chelsea is at Aston Villa in another match Saturday, when third-place Arsenal is at Queens Park Rangers.

Everton hosts West Bromwich Albion, Fulham hosts Norwich, Wigan hosts Stoke and Wolverhampton Wanderers hosts Bolton.

On Sunday, it's Tottenham vs. Swansea and Newcastle vs. Liverpool.

IPO market heats up amid cautious optimism

NEW YORK (AP) ? A better economic outlook and rising stock market are heating up the market for initial public stock offerings this spring, just weeks before Facebook's highly anticipated IPO.

In the past two days, two companies have posted the biggest first-day gains for a freshly public company since LinkedIn Corp., the professional networking service, had its IPO last May.

Congress, meanwhile, is lessening restrictions on IPOs in an attempt to make it easier for younger companies to raise money. Still, with ongoing worries about high gas prices and unemployment dampening the economy, it's too early to tell whether an IPO recovery is here to stay.

The willingness of more startups to take the plunge suggests that they are optimistic about their business, and also the broader economy. And a strong IPO market could mean more jobs, as companies loaded with fresh cash from investors hire on workers to grow their operations.

This week, shares of Berkeley, Calif.-based Annie's Inc., best known for its organic mac n' cheese and bunny-shaped crackers, soared 89 percent on Wednesday. On Thursday, shares of Baltimore mobile advertising company Millennial Media Inc. jumped 92 percent. Ten companies are expected to go public this week, which would be the most since December 2010.

That comes on top of a strong quarter for IPOs. So far this year, the average first-day pop is around 13 percent, according to the IPO advisory firm Renaissance Capital, and that's the best since before the financial crisis and global economic meltdown. There have been more IPOs in the first three months of 2012 than there have been since the April-June quarter of last year, said Richard Petersen, credit analyst at S&P Capital IQ.

The rush is coming after the economy grew at an annual rate of 3 percent final three months of 2011, the best pace in a year and a half, and hiring is picking up. The Standard & Poor's 500 index closed at its highest point since May 2008 earlier this month. An improving stock market helps fuel demand for IPOs, which are considered riskier investments.

But the IPO market may yet peter out. After all, 2011 started out with signs for optimism, but the European debt crisis and renewed economic weakness put a damper on the second half of the year, freezing the IPO market for months. And while more companies are going public now, they're pulling in less cash. So far this year, U.S. IPOs have raised $5.5 billion, well below the $13 billion as of the same time last year, according to Renaissance Capital. It's also still too early to tell whether efforts by lawmakers to make it easier for small companies to raise money through IPOs will help rev up the market.

The big first-day pops and slew of companies launching IPOs are drawing big headlines, but what matters for investors is what the company does beyond that first day, said Kathleen Smith, principal at Renaissance Capital.

Small-time investors could even lose money if they buy into companies during the first-day madness when the stock is trading well above the price that big investors, such as hedge funds and mutual funds, got the night before.

"A more thoughtful investor might wait until the frenzy is over," she said.

LinkedIn, for example, more than doubled in its May debut, closing up 109 percent at $94.25. But the hubbub didn't last, and LinkedIn, though profitable, saw volatility that's typical with newly traded stocks. It currently hovers around $100 after steady gains this year.

Though far from the dot-com bubble of the late 1990s, much of the investor demand in the past year has centered on technology stocks like LinkedIn, especially companies engaged in mobile devices and social networking.

Demand for technology companies is at a fever pitch with Facebook's IPO, expected later this spring. The Menlo Park, Calif.-based company has said it anticipates raising $5 billion in its IPO, but that amount could double by the time it files its final paperwork. If all goes as expected, world's biggest online social network could be valued at as much as $100 billion once it goes public. That would be the biggest Internet IPO ever.

S&P's Petersen said the "Facebook factor" and gains in the broader stock market are both helping stoke investor demand for IPOs. It's also the end of the first quarter, so it's not uncommon for banks to want to get deals in before the period closes, he said.

There are about 200 companies waiting to go public right now, he said, adding that the question will be whether investors buy in.

Santorum beats Romney in Louisiana

His victory in Louisiana gives Rick Santorum bragging rights, but it does not change the overall dynamics of the race. He still dramatically lags behind Mitt Romney in the hunt for delegates.

Rick Santorum won the Louisiana Republican presidential primary Saturday, beating front-runner Mitt Romney in yet another conservative Southern state.

Although the victory gives Santorum bragging rights, it does not change the overall dynamics of the race; the former Pennsylvania senator still dramatically lags behind Romney in the hunt for delegates to the GOP's summertime nominating convention.

Even so, Santorum's win underscores a pattern in the drawn-out race.

The under-funded under-dog has tended to win in Bible Belt states that include Tennessee, Mississippi and Alabama. Romney ? a deep-pocketed, highly organized former Massachusetts governor ? has persistently struggled in such heavily conservative regions.

Neither candidate was in the state as Louisiana Republicans weighed in. Nor was former House Speaker Newt Gingrich, who was trailing in Louisiana.

Romney took a rare day off Saturday, with no public events. Santorum spent the day campaigning in Pennsylvania and next-up Wisconsin, which votes April 3 and represents one of his last chances to beat Romney in a Midwestern state.

Early exit polls conducted for The Associated Press and the television networks showed that Santorum's win in Louisiana was one of his strongest performances to date among conservatives, working class voters, and those calling the economy their top issue. And he continued his dominance among white evangelical voters and those looking for a candidate who shares their religious beliefs.

As in previous Southern states, Romney's best showing came among those voters with annual incomes above $100,000 and those who prioritized a candidate's ability to defeat President Barack Obama in November.

The bad economy was the top issue for Louisiana voters. Most were gloomy about prospects for a recovery, saying they felt the economy was getting worse instead of better. While some national surveys suggest Americans are feeling optimistic about economic improvement, just one in eight Republican primary voters said they thought a recovery was under way.

Santorum badly needed a rebound after a decisive Illinois loss to Romney earlier in the week that moved party stalwarts to rally around the front-runner. Many urged Santorum and Gingrich to drop out of the race.

Justin Berrier: Krauthammer Brings Thoroughly Debunked Health Care Cost Myth To The Washington Post

In his March 22 Washington Post column, conservative commentator and frequent health care misinformation peddler Charles Krauthammer parroted the latest right-wing myth?about the Affordable Care Act (ACA): that a recent Congressional Budget Office (CBO) estimate of the cost of the ACA showed that the law will cost twice as much as a similar estimate from two years ago. From The Washington Post?(emphasis added):

Obamacare was carefully constructed to manipulate the standard 10-year cost projections of the CBO. Because benefits would not fully kick in for four years, President Obama could trumpet 10-year gross costs of less than $1 trillion -- $938 billion to be exact.

But now that the near-costless years 2010 and 2011 have elapsed, the true 10-year price tag comes into focus. From 2013 through 2022, the CBO reports,?the costs of Obamacare come to $1.76 trillion?-- almost twice the phony original number.

Rarely has one law so exemplified the worst of the Leviathan state -- grotesque cost, questionable constitutionality and arbitrary bureaucratic coerciveness. Little wonder the president?barely mentioned?it in his latest?State of the Union address. He wants to be reelected. He'd rather talk about other things.

Of course, the claim that the cost of the health care law has suddenly doubled has been thoroughly debunked over the past week by numerous sources -- for example, here, here, here, and here. Krauthammer can't plead ignorance, as even his Washington Post colleagues have shot down this myth. In a March 20 post on his Washington Post blog, Wonkblog, Ezra Klein provided this helpful chart -- two days before Krauthammer weighed in -- explaining that an "apples-to-apples graph of the 'net' cost of the coverage provisions" showed a decrease in the cost of the law:

The title of Klein's post makes the truth plainly clear: "No, the CBO hasn't doubled its cost estimate for health-care reform." Again, this was published on The Washington Post's website two days before Krauthammer's column went up.

Although largely dedicated to perpetuating this myth, even a few members of the right-wing media have admitted that the claim is a distortion of the CBO's report. On the National Review Online blog The Corner, Patrick Brennan, who describes the law as a "Brobdingnagian bureaucracy" with "fishy accounting" explained:

This claim, that the CBO's 2012 estimate suggests Obamacare will cost twice as much as originally projected when the bill was passed in 2010, has been widely trumpeted, by some?rather doggedly, as another Obamacare failure, but unfortunately, it's entirely dishonest accounting, as a range of liberal bloggers have pointed out.

Here's why: The gross costs of Obamacare's insurance coverage in the CBO's 2010-2019 estimate were indeed $940 billion (table 2, page 2,?here). The updated estimate covers the years 2012-2022, in which the gross costs will be $1.76 trillion, which is, yes, almost double the other number (table 2,?here). But these numbers aren't remotely comparable.

Pretending that the $940 billion number has been revised to $1.76 trillion is like comparing Tom Brady's first through eighth seasons in the NFL with his third through eleventh (though probably only Paul Krugman thinks of?government?spending?as a touchdown pass). Gross costs in each year haven't really changed -- e.g., in 2019, the old, 2010 estimate predicted a gross cost of $216 billion, the new estimate is $224 billion (the net costs of coverage have consistently decreased from the old estimate to the new -- in 2019, say, from $175 billion to $149 billion).

Given Krauthammer's history of pushing conservative myths about the health care law, perhaps it's not surprising he would highlight this latest falsehood. What is surprising, though, is The Washington Post publishing it after already debunking it.

International Medical University to hold cycling event in district

BATU PAHAT: The International Medical University (IMU) will be holding a cycling event in conjunction with its 20th Anniversary celebration.

Members of the public are invited to participate in a 4-km leisure ride in town and a 25km road race at the outskirts of Batu Pahat on April 14.

Event organiser Canace Teoh said a participation fee of RM30 would be charged for cyclists taking part in the male and female category road race.

?All participants must be 18 years of age and we will flag off the event Jalan Minyak Beku at 7am on the race day.? She added that cash prizes awaits the top three cyclists.

For the leisure ride, cyclists taking part in the event would be flagged off at Dataran Penggaram in Batu Pahat on the same day.

NASA considering space station for Mars dry run

CAPE CANAVERAL, Fla. (AP) ? The International Space Station may provide the setting for a 500-day pretend trip to Mars in another few years.

NASA said Tuesday that consideration is under way to use the space station as a dry run for a simulated trip to and from Mars.

It would be patterned after Russia's mock flight to Mars that lasted 520 days at a Moscow research center. Six men were involved in that study, which ended late last year. They were locked in a steel capsule.

NASA's space station program manager Mike Suffredini said before astronauts can fly beyond low-Earth orbit, they'll have to spend more than six months aloft at a time. That's the typical stint for space station crews. Five hundred days is more than 16 months.

The human endurance record of 14 months was set by a Russian cosmonaut aboard the Mir space station in the mid-1990s. Only two others ? both Russians ? have spent as long as a full year in space.

No NASA astronaut has spent more than seven months in space on a single mission.

Suffredini doesn't expect any such Mars simulation aboard the space station to occur any sooner than two to three years. Physical as well as psychological questions will have to be addressed before anything of that sort is attempted, he said.

The diagnosis of myelodysplastic syndrome, a blood cancer, often causes confusion. While some patients can be treated with repeated blood transfusions, others require chemotherapy, leaving some uncertainty about whether the syndromes actually are cancer.

Now, using the latest DNA sequencing technology, scientists at the Washington University School of Medicine in St. Louis have shown that the blood disease is an early form of cancer with characteristics that are very similar to the fatal leukemia to which it often progresses. And by mapping the genetic evolution of cancer cells in seven patients with myelodysplastic syndromes who later died of leukemia, they have found clues to suggest that targeted cancer drugs should be aimed at mutations that develop early in the disease.

The research, by a large team of Washington University researchers at the Siteman Cancer Center, appears online March 14 in the New England Journal of Medicine.

The scientists sequenced all the DNA ? the genome ? of tumor cells from the patients over time. While some cancer cells in each patient acquired new mutations as they evolved, they always retained the original cluster of mutations that made the cells cancerous in the first place.

This discovery, which must be confirmed in larger studies, suggests that drugs targeted to cancer mutations might be more effective if they are directed toward genetic changes in the original cluster of cancer cells called the founding clone. Drugs that target mutations found exclusively in later-evolving cancer cells may kill those cells but likely wouldn't damage founding clones that do not carry the later mutations.

"It's probably not enough to know that a particular mutation exists in cancer cells," says senior author Timothy Graubert, MD, associate professor of medicine at the School of Medicine who also treats patients at Barnes-Jewish Hospital. "We likely will need to dig deeper to find out whether a mutation is in the founding clone that initiated the cancer or in a later-evolving clone."

In other words, think of this cancer as a tree, Graubert says.

"To kill a tree, you have to pull out the roots," he says. "If you only cut off a limb, it will just grow back. We're saying that to be effective, targeted cancer drugs probably need to attack mutations at the root of this disease."

About 28,000 Americans are diagnosed with myelodysplastic syndromes each year, most over age 60. They occur when blood cells produced in the bone marrow don't fully develop and immature cells crowd out healthy ones. In about one-third of patients, the disease progresses to a fatal form of leukemia.

As part of the new research, Graubert and his colleagues teamed with researchers at Washington University's Genome Institute who sequenced the genomes of cancer cells after the patients developed acute myeloid leukemia. Then, they determined whether the mutations they found were present when the same patients were first diagnosed with myelodysplastic syndromes.

They identified every mutation, typically hundreds, that developed in each patient's bone marrow cells as the cancer evolved. They also showed that about 85 percent of the patients' bone marrow cells were cancerous, regardless of whether they had myelodysplastic syndromes or leukemia.

Even in the earliest stages of myelodysplastic syndromes, when typically only a small number of immature blood cells populate the bone marrow, roughly 85 percent of bone marrow cells were part of the malignant clone.

"These results clearly establish that myelodysplastic syndromes are truly an early form of cancer," says first author Matthew Walter, MD, assistant professor of medicine, who also treats patients at Barnes-Jewish Hospital. "But until now, there were a lot of people ? patients and physicians included ? who questioned this."

That such a high percentage of bone marrow cells are malignant so early in the course of myelodysplastic syndromes that progress to leukemia may help improve the diagnosis of the disease and aid in determining prognosis, Walter says.

In the current study, funded in part by a federal stimulus grant from the National Institutes of Health (NIH), the researchers also identified 11 mutations in the patients' cancer cells that were later found to occur in other patients with acute myeloid leukemia, an indicator of the mutations' significance. Four of these mutations had never before been linked to myelodysplastic syndromes or leukemia.

To track the evolution of cancer cells, the researchers captured segments of DNA involved in every mutation and repeatedly sequence those regions more than 600 times each. Using this deep sequencing approach, developed at The Genome Institute, they could identify not only the founding clone in each patients' bone marrow cells, but also "breakaway" secondary clones that contributed to both the progression of myelodysplastic syndromes and acute leukemia. In all cases, the secondary clones could be traced back to the founding clone.

"This tells us that the secondary clones were not distinct cancers, but that they all evolved from the founding clone," Walter says.

The researchers say that sequencing the entire genomes of the cancer cells was essential to piecing together a picture of the way cancer evolved. While this technology is not yet routinely available to cancer patients, Graubert and Walter say reduced sequencing costs and improved analytical approaches should make it easier for more scientists to get a sense of the clonal nature of a patient's tumor cells.

The President of Afghanistan is asking the U.S. to hand over all security responsibilities by 2013. NBC's Richard Engel reports.

Updated at 3:45 p.m. ET: Afghanistan's president on Thursday called for U.S. and other foreign forces in Afghanistan to leave villages in the country and move to larger bases instead, according to Hamid Karzai's office.

In a statement -- which comes amid the ongoing controversy over the killing of 16 Afghan civilians in Panjwai, Kandahar province,?allegedly by a lone U.S. soldier -- Karzai's office?said?he had made the request to U.S. Defense Secretary Leon Panetta and said that "Afghan Security forces currently have the ability to secure the villages around the country."

The?statement also said that NATO-led foreign?forces needed to have "complete respect for their religion and the Afghan culture. No foreign troops should enter Afghans homes. And they should pay more attention on reconstruction and financial support for our country."

The American soldier accused of gunning down 16 Afghan civilians, including nine children, has been flown out of Afghanistan to Kuwait. NBC's Richard Engel reports.

"Karzai said that both sides should work on accelerating the process and that they should hand over all the security responsibilities in 2013 instead of 2014," it added.

"We demand that the process move quickly and they transfer authority into Afghan hands," it added.

In a near-simultaneous announcement, the Afghan Taliban said it was suspending nascent peace talks with the United States seen as a strong chance to end the country's decade-long conflict, blaming "shaky, erratic and vague" U.S. statements.

The White House denied Taliban accusations?and insisted that Washington had been consistent in its message.

"We support an Afghan-led process toward reconciliation. There is no likely resolution to the conflict in Afghanistan without a political resolution," White House spokesman Jay Carney told reporters.

At the State Department, spokeswoman Victoria Nuland said the U.S. is still "prepared to continue these discussions," but will "have to see where this goes now," NBC News reported. "We still feel that if there is a process that can be supported that we ought to do that ... Our only goal is to get Afghans to sit down together," she said.

Nuland said the process so far has had a "lot of ups and downs."

Gavin Sundwall, U.S. Embassy spokesman in Kabul, echoed the comments, NBC News said.

On Karzai?s statement, he also said, ?The transition to Afghan security lead is already under way,? NBC News reported. ?About half of all Afghans currently live in areas where Afghan security forces are taking responsibility.?

He said NATO will decide the next phase of transition including the shift of coalition forces to a support role in 2013.

'Cruel and brutal'
The announcement from Karzai's office said Panetta had "once again paid his condolences to the Afghan President and to the people of Afghanistan on the behalf of his government and his people for the Panjwai incident."

Karzai described the killings of the 16 people, including women and children, as "a cruel and brutal action."

The soldier accused of carrying out the Panjwai shooting was based at a small special forces compound similar to others around the country that underpin NATO's counter-insurgency strategy.

"I assured him first and foremost that I shared his regrets about what took place. I again pledged to him that we are proceeding with a full investigation here and that we will bring the individual involved to justice. He accepted that," Panetta told reporters before leaving Afghanistan.

It appears highly unlikely that the staff sergeant accused of killing 16 Afghan civilians will be returned to Afghanistan for court martial. NBC's Jim Miklaszewski reports.

The Afghan Taliban, which describes itself as the Islamic Emirate of Afghanistan,?said it had decided to suspend peace talks after opening a diplomatic office in Qatar earlier this year to enable talks to go ahead.

"It seems that the invading Americans and their stooge regime took advantage of these measures of Islamic Emirate and sought to achieve other malicious objectives and therefore are postponing the core issues and are wasting time," the Taliban said.

"So the Islamic Emirate has decided to suspend all talks with Americans taking place in Qatar from today onwards until the Americans clarify their stance on the issues concerned and until they show willingness in carrying out their promises instead of wasting time," it added.

The austere Islamist movement said it was fully prepared to continue its "long-term Jihadi strategies" as the traditional summer fighting months approached, following a harsh winter which had dulled fighting in several volatile provinces.

In the latest attack, a roadside bomb killed 13 Afghan civilians, including women and children, and wounded two on Thursday in the south of the country, provincial officials said.

More than 3,000 civilians were killed in the war in Afghanistan in 2011, the fifth year in a row the number has risen, according to the United Nations.
